利用PCR分别合成掺入地高辛标记的35S启动子和NOS终止子核酸探针,再利用放射免疫技术将I^125标记的地高辛抗体与35S启动子和NOS终止子探针分别进行杂交检测。结果显示转基因样本的杂交信号均为阳性,且灵敏度较高。特异性强,为转基因植物的分析检测又提供了一种较实用的方法。 相似文献

通过体外和体内试验研究了改性坡缕石对肉鸭日粮中黄曲霉毒素B1的去毒作用.体外试验表明与坡缕石相比,改性坡缕石对黄曲霉毒素B1的吸附率更强.480羽1日龄樱桃谷商品代肉鸭,按饲养试验要求随机分为4组1)对照组为基础日粮;2)基础日粮+0.25%改性坡缕石;3)霉变玉米50%代替基础日粮中的玉米,使黄曲霉毒素B1前期为50μg/kg,后期为55μg/kg;4)第3组日粮+0.25%改性坡缕石.结果表明与对照组相比,添加0.25%改性坡缕石对肉鸭生长性能、组织和血清黄曲霉毒素B1含量、内脏器官相对质量、血清生化指标均无显著影响.然而,霉变玉米使肉鸭生长性能显著降低,使胸肌、肝脏、肾脏、血清黄曲霉毒素B1含量以及肝脏率、肾脏率、胰脏率、血清谷草转氨酶、谷丙转氨酶、淀粉酶水平显著提高;使血清总蛋白、白蛋白、尿酸水平显著降低.联合饲喂霉变玉米/改性坡缕石可使肉鸭生长性能、组织器官和血清黄曲霉毒素B1含量、内脏器官相对质量、血清生化指标均恢复至对照组水平.上述结果提示,改性坡缕石对肉鸭日粮中黄曲霉毒素B1具有较好的去毒作用. 相似文献

The impact of biocontrol strain Pseudomonas fluorescens CHA0 and of its genetically modified, antibiotic-overproducing derivative CHA0/pME3424 on a reconstructed population of the
plant-beneficial Sinorhizobium
meliloti bacteria was assessed in gnotobiotic systems. In sterile soil, the final density of the reconstructed S. meliloti population decreased by more than one order of magnitude in the presence of either of the Pseudomonas strains when compared to a control without addition of P. fluorescens. Moreover, there was a change in the proportion of each individual S. meliloti strain within the population. Plant tests also revealed changes in the nodulating S. meliloti population in the presence of strains CHA0 or CHA0/pME3424. In both treatments one S. meliloti strain, f43, was significantly reduced in its root nodule occupancy. Analysis of alfalfa yields showed a slight but statistically

Using regression analysis method,the methods for solving the weights of combination forecasting model(CFM) are proposed. At first, the linear regress CFM are presented based on the least absolute criteria and least square criteria. Then the weights can be evaluated using the least square princinple. Because the objective function of CFM based on least absolute criteria is non differential, the traditional programming methods can not solve it. So the least square method with the modified weights is proposed to solve this problem. At the same time, methods for solving CFM is given with the aim of minimizing sum of percentage error absolutes. The study aimed to explore the intestinal microbial community changes after transfering the IGF-1 gene into the superfine wool sheep, which would lead to potential problems in the biosafety of transgenic sheep. Forty one individuals in IGF-1 transgene positive group (GP group, female(GPF) 24 and male(GPM) 17), 43 individuals in genetically modified negative group (GN group, female(GNF) 25 and male(GNM) 18) and 29 individuals in non-genetically modified group(NG group, female(NGF) 18 and male(NGM) 11) from clinical healthy sheep in the same field were randomly selected. The rectal fecal samples were collected. The Ⅱlumina HiSeq high-throughput sequencing technology was used to determine bacterial 16S rRNA V3-V4 area sequence, and the bacteria community composition in fecal samples of transgenic superfine wool sheep and non-transgenic sheep was analyzed. There were 17 phyla, 32 classes, 56 orders, 94 families, 228 genera and 185 species. LEfSe analysis showed that there were 10 biomarkers in the GPF group, there were 5 biomarkers in GPM group, all of which were common colonization bacteria of ruminant intestinal flora. The biomarkers in NGF group was Bacteroides BS11 family, the biomarkers in NGM group was Firmicutes. They were the main dominant bacteria of ruminant intestinal flora. Analysis of shared bacteria of intestinal flora in 3 groups showed that the proportion of shared bacteria was as high as 91%-94% in 6 taxa of genus and family. It was confirmed that the introduction of IGF-1 gene did not change the overall distribution of intestinal flora in superfine wool sheep, and the transgenic sheep were biosafety and environmental safety. 相似文献
